Plasmid vector

Abstract

PURPOSE:To obtain a plasmid vector suitably useful as a shuttle vector for Bifidobacterium longum, by bonding plasmid pBR322 derived from Escherichia coli to a plasmid derived from Bifidobacterium longum. CONSTITUTION:Plasmid vector pBR derived from Escherichia coli is integrated with plasmid (e.g. pTB4 or pTB10) derived from Bifidobacterium longum and optionally with the whole of plasmid pC194 derived from Staphylococcus aureus or a chloramphenicol-resistant gene range thereof and/or tryptophan synthetic gene to give plasmid vector. For example, plasmid vector such as pTBR101 wherein pBR322 is bonded to pTB4 at BamHI site, pTBR102 wherein pBR322 is bonded to pTB10 at BamHI site, pTBR101-CM wherein chloramphenicol- resistant gene of pC194 is further bonded to pTBR101, etc., may be cited as the plasmid vector.
